Mashisu Makibi showed up as Misao Kuramitsu's second-in-command aboard the Choubimaru in the third Tenchi OVA, though she was actually a last-minute fill-in when Mikami Kuramitsu replaced Noike Kamiki Jurai in the role. The Choubimaru was a planet-destroyer-class ship, and Mashisu spent most of her time trying to keep Misao from doing something catastrophically stupid.
Misao's plan was nasty: rescue his sister Mihoshi from Tenchi using the ship, then "accidentally" let the main battery malfunction during a fight with Ryoko, which would wipe out Earth and kill Tenchi along with it. The whole thing was actually Z's manipulation—she'd forged the letter about Mihoshi being in danger—but Misao didn't know that. Mashisu begged him to change course, even resorting to spanking him to save him from the embarrassment of showing up to crew meetings with a black eye. When reasoning failed, she came up with her own plan: frame Ryoko for destroying the ship instead.
That backup plan fell apart fast. Sasami beat Mashisu in combat and also clued her in that—oh yeah, you're in love with Misao. By the time Mashisu got back to the Choubimaru after a drunken Ryoko started wrecking things, Misao had finally snapped out of it. But when an explosion hit during the evacuation, Misao threw himself in front of Mashisu to shield her.
Everyone thought she was done for. She even confessed her love while apparently dying, and had a whole emotional flashback to when they first met—Misao had nailed her in the head with a tennis ball, then invited her to lunch with his family afterward. Except the whole near-death thing was fake: Washu had rigged an illusion to make Misao understand what actually mattered. Mashisu was mortified that everyone had heard her confession.
The fallout was rough. Mikami sentenced her to six months cleaning toilets at Galaxy Police headquarters. Seto stepped in and adopted Mashisu so she could marry Misao, which Mitoto—Misao and Mihoshi's mom—showed up to confirm was approved by the Kuramitsu Family. They actually went through with it, and Mashisu got the Choubimaru as a wedding present while Misao joined the Galaxy Army.
Later on, in GXP episode 15, Mashisu was back in action helping Fuku when the Kamidake II got attacked by Daluma guild pirates. She and Ryo-Ohki chased them off together.
