Mitsuishi graduated high school in 1986 and enrolled at Katsuta Seiyū Academy. To support herself while studying, she worked part-time as an elevator operator at the Sunshine 60 building, then briefly as an office lady—though she couldn't stick with the latter job due to frequent absences.
Her voice acting debut came in 1989 with a minor role in the OVA Ace o Nerae! Final Stage, but she became a household name three years later as Usagi Tsukino in Sailor Moon. That role catapulted her to stardom, and she cemented her status as one of the industry's most influential seiyū when she voiced Misato Katsuragi in Neon Genesis Evangelion. Her pull in the industry is serious enough that the anime adaptation of Ebichu was greenlit largely because of her personal interest in bringing it to life.
These days she's a freelancer (formerly with talent agency Arts Vision) and one of the few major seiyū who actually rides a motorcycle—a Yamaha FZ250 Phaser, a fun callback to those old office lady days. She's married with one daughter.
